The iMSRC graphical interface is accessible through a web page where the user can easily choose among different options, allowing fine-tuning of all parameters necessary for capturing and detection. The parameters required to carry out the entire process include:
- settings for capturing the searching mosaic. They should be saved as proprietary software capturing files and kept in a shared folder, and contain the parameters to carry out the first step. They are set in the “Select Low Resolution Scan Settings” field.
- image analysis routines for object detection within the searching mosaic image to perform the second step. It is set in the “Select objects detection routine” button and is possible to choose from different analysis routines previously saved, and to adjust the variables in the “Advance Options” button.
- second-scan acquisition settings files. They should be saved as proprietary software capturing files and kept in a shared folder, like the settings required for the first step, and contain the parameters to carry out the third step. They are set in the “Select template 2” field.
In order to launch the fully automated acquisition, the microscope should be equipped with a communications port (i.e. the Computer Aided Microscope port [CAM] of Leica microscopes), so iMSRC can automatically launch the whole process that directs information flow between the capturing and analysis software by means of a single click (“Play” button).
